Therapeutic Applications of Azanucleoside Analogs as DNA Demethylating Agents

چکیده
Azanucleosides, such as 5-azacytidine and decitabine, are DNA demethylating agents used in the treatment of acute myeloid leukemia myelodysplastic syndromes. Researchers continue to explore their utility other hematologic solid tumors. Based on capacity compounds inhibit methyltransferase enzymes important role methylation health disease, it is essential understand molecular changes that azanucleosides induce how these may improve outcomes subsets patients. This review summarizes therapeutic actions discusses recent clinical trials single or combination therapy for cancer related conditions.

DNA demethylating agents synergize with oncolytic HSV1 against malignant gliomas.
PURPOSE Oncolytic viruses (OV) based on herpes simplex virus type 1 (HSV1) are being used in clinical trials for a variety of cancers. The OV, rQNestin34.5, uses a nestin promoter/enhancer to selectively drive robust viral replication in malignant glioma cells. We have discovered that this promoter becomes extensively methylated in infected glioma cells, reducing OV efficacy. EXPERIMENTAL DES...
متن کاملClinical development of demethylating agents in hematology.
The term epigenetics refers to the heritable changes in gene expression that are not associated with a change in the actual DNA sequence. Epigenetic dysregulation is linked to the pathogenesis of a number of malignancies and has been studied extensively in myelodysplastic syndromes and acute myeloid leukemia. DNA methylation is frequently altered in cancerous cells and likely results in transcr...
متن کاملDemethylating Agents in the Treatment of Cancer
Gene silencing resulting from aberrant DNA methylation can lead to tumorigenesis. Therefore, drugs that inhibit or interfere with DNA methylation have been used to reactivate and induce silenced gene re-expression in malignancies. Two demethylating agents, azacitidine and decitabine, are approved for the treatment of myelodysplastic syndromes (MDS) by the U.S. Food and Drug Administration (FDA)...
متن کاملMushrooms as therapeutic agents
Mushrooms have been known for their nutritional and culinary values and used as medicines and tonics by humans for ages. In modern terms, they can be considered as functional foods which can provide health benefits beyond the traditional nutrients. There are monographs that cover the medicinal and healing properties of some individual traditional mushrooms. There has been a recent upsurge of in...

A relatively noncytopathic virus, lymphocytic choriomeningitis virus (LCMV),1 prevents the development of insulin-dependent diabetes mellitus (IDDM) in nonobese insulin-dependent diabetes (NOD) mice (1, 2). Typically, >90% of NOD female mice, by 9 mo of age, develop hyperglycemia, hypoinsulinemia, and lymphocytic infiltrates in the islets of Langerhans, with a cell destruction . However, their ...
